稀土摻雜上轉(zhuǎn)換氟化物納米材料的生物應(yīng)用(標(biāo)記及探針)
1:稀土摻雜上轉(zhuǎn)換氟化物納米顆粒在生物分子檢測中的應(yīng)用
利用靜電吸引層層組裝的方法在NaYF4BYb3+,Er3+外部引入氨基,從而能夠和生物素相連,隨后體系中再加入同生物素相連的金納米顆粒,當(dāng)抗生物素蛋白存在的時候會連接兩種納米顆粒,從而發(fā)生能量轉(zhuǎn)移,此時金會吸收稀土上轉(zhuǎn)換納米材料的熒光,通過熒光猝滅的程度實現(xiàn)抗生物素蛋白的檢測,檢測范圍為0.5~370mol/L。
氨基修飾的上轉(zhuǎn)換納米顆粒不僅可以和蛋白偶聯(lián),還可以和DNA、小分子相連,將上轉(zhuǎn)換納
米顆粒結(jié)合磁場生物分離技術(shù)用于微量DNA的檢測。非常巧妙的利用Lemieux2vonRudloff試劑直接氧化合成過程中引入的油酸配體,得到羧基修飾的上轉(zhuǎn)換納米顆粒,用于定量檢測靶向DNA。將Fe3O4和NaYF4BYb,Er納米顆粒包覆在二氧化硅微球內(nèi),制備的發(fā)光磁性多功能復(fù)合納米顆粒,在生物分離、檢測、標(biāo)記成像及藥物輸送方面有著潛在的應(yīng)用價值。
2:稀土摻雜上轉(zhuǎn)換氟化物納米在免疫分析中的應(yīng)用
2001年,Niedbala課題組報道了上轉(zhuǎn)換納米顆粒應(yīng)用于免疫層析實驗。研究者將上轉(zhuǎn)換材料同生物分子相連后,采用免疫層析技術(shù)對抗原進行檢測取得了理想的結(jié)果。由于層析所用的底板在紅外光照射下不發(fā)光,所以觀測到的信號必然來自上轉(zhuǎn)換納米顆粒,因此上轉(zhuǎn)換納米材料用于免疫層析技術(shù)更加提高了這種方法的可靠性。400nm的UCNP與Cy5相比,目標(biāo)物濃度和發(fā)光強度的線性關(guān)系提高兩個數(shù)量級。此外,研究者將膠體金和上轉(zhuǎn)換納米顆粒同時應(yīng)用于HPV16的檢測,結(jié)果表明UCNP實驗的檢測限低于膠體金的100倍。但需要說明的是,控制上轉(zhuǎn)換納米顆粒的尺寸是擴展其在免疫層析技術(shù)應(yīng)用的關(guān)鍵因素。
3:稀土摻雜上轉(zhuǎn)換氟化物納米顆粒在生物標(biāo)記中的應(yīng)用
稀土上轉(zhuǎn)換氟化物納米顆粒在生物標(biāo)記中的應(yīng)用成為當(dāng)前納米標(biāo)記的熱點。最初的活體實驗是用線蟲吞噬稀土上轉(zhuǎn)換氧化物納米顆粒。制備了氨基修飾的摻雜FITC的上轉(zhuǎn)換二氧化硅納米顆粒,又將能與癌細胞特異性結(jié)合的葉酸共價鍵連接在納米顆粒外面。這種納米顆??梢噪p模式上轉(zhuǎn)換下轉(zhuǎn)換發(fā)光,同時實現(xiàn)對癌細胞的標(biāo)記,用于流式細胞儀進行分選,將葉酸修飾到聚乙烯亞胺包覆的NaYF4BYb,Er納米顆粒上,進行了一系列的細胞以及白鼠活體實驗。研究表明此納米顆粒在磷酸鹽緩沖溶液中非常穩(wěn)定,同時對骨髓干細胞沒有毒性,抗漂白性好。將其應(yīng)用于人卵巢癌細胞和結(jié)腸癌細胞細胞標(biāo)記,修飾后的納米顆粒和細胞表面具有較高的親和力。小白鼠實驗中,將UCNP和量子點同時應(yīng)用于皮下組織成像,在紫外光照射下腹部皮下組織的量子點沒有光信號,而在980nm的激發(fā)下,可以清楚看到UCNP的發(fā)射光,證實了UCNP在生物活體標(biāo)記中的潛在價值。
4:稀土摻雜上轉(zhuǎn)換氟化物納米在光動力治療中的應(yīng)用
第一次將稀土上轉(zhuǎn)換納米顆粒作為光敏劑的能量供體,包覆二氧化硅的同時摻雜部花青,實現(xiàn)UCNP和部花青的能量轉(zhuǎn)移產(chǎn)生單線態(tài)氧。將約50nm大小的共價連接葉酸、非共價吸附酞菁鋅的聚乙烯亞胺包覆的NaYF4BYb,Er納米顆粒,應(yīng)用于活體實驗。酞箐鋅的吸收波長和NaYF4:Yb,Er的670nm處的發(fā)射有重疊,發(fā)生能量轉(zhuǎn)移,當(dāng)用980nm的激發(fā)上轉(zhuǎn)換納米顆粒時,光敏劑會產(chǎn)生單線態(tài)氧。將這種多功能的納米顆粒應(yīng)用于活體實驗,證明了此種納米顆粒不僅細胞毒性小,而且可以靶向識別癌細胞,并且可以有效的殺死腫瘤細胞,癌細胞殺死率高達80%。
