【熒光標(biāo)記人/牛血清白蛋白】:

Bovine serum albumin (BSA) is a slightlyacidic ~66 kd protein. BSA is commonly used in biological research due to itslow cost and inertness. Dilute enzymes can be stabilized by BSA (useful forrestriction digests). BSA is a common blocking agent in western blotting andELISA’s. Although, bovine serum albumin is exceptionally soluble (up to 50% w/vin buffered saline), it is typically used at a concentration of 0.1-1% w/v.

牛血清白蛋白,英文名稱為BovineSerum Albumin,也稱Bovinealbumin,或CohnFraction V,簡稱BSA。BSA有著極其廣泛的實驗室應(yīng)用,在免疫實驗中常用作封閉劑,包括ELISAWBWestern Blot)。作為載體蛋白,將其交聯(lián)于半抗原和其他弱抗原可以使它們在抗體生產(chǎn)中具有更強的免疫原性。在限制性內(nèi)切酶消化反應(yīng)中,BSA常常被用作一些酶的反應(yīng)穩(wěn)定劑,并且能防止它粘附到管壁和槍頭上。BSA也常用于生物制藥的生產(chǎn)過程或者作為營養(yǎng)物用于細(xì)胞和微生物培養(yǎng)。除此之外,還作為蛋白定量檢測的標(biāo)準(zhǔn)品使用。我們提供的BSA產(chǎn)品,使用優(yōu)質(zhì)牛血漿,利用熱休克法(Heat Shock)法制備而成。

 

牛血清白蛋和人血清白蛋白在藥物靶向和藥物傳遞方面的介紹:

與正常組織相比,腫瘤組織的血管豐富,結(jié)構(gòu)特殊 一個重要的表現(xiàn)就是血管內(nèi)皮間隙較大,這種病變情況下大分子可以透過腫瘤組織血管壁的原因,然而在健康組織的血管中只有小分子可以通過這一內(nèi)皮屏障,通常大分子藥物流體動力學(xué)半徑大于2nm小于10nm 人血清白蛋白的有效半徑是7.2nm 因此可以滲入腫瘤組織而不能進(jìn)入正常組織。

在病例條件下 腫瘤細(xì)胞在生長或轉(zhuǎn)移時,病變細(xì)胞釋放的化學(xué)物質(zhì)和腫瘤衍生物的甲狀腺激素相關(guān)肽均通過產(chǎn)生NO和激活KATP來降低集合淋巴管“節(jié)律泵”的活性,從而影響了淋巴的生成 轉(zhuǎn)運使得腫瘤組織淋巴回流較小,分子量相對較小的物質(zhì)從腫瘤間質(zhì)中清除較快,而大分子被截留,因此大分子物質(zhì)還可以在腫瘤組織中選擇性的聚集。高血管通透性和截留率是白蛋白作為抗腫瘤藥物靶向載體的必要條件,白蛋白的這種效應(yīng)符合大分子物質(zhì)在腫瘤組織的透過性增強及滯留效應(yīng)。

【熒光標(biāo)記和功能化卵清蛋白】

蛋清中的主要蛋白質(zhì)包括卵清蛋白、卵轉(zhuǎn)鐵球蛋白、類卵沾蛋白、卵沾蛋白、溶菌酶、球蛋白G2、球蛋白G3等。卵清蛋白等電點4.5,其本質(zhì)是含磷糖蛋白。卵清蛋白是蛋清中的主要蛋白組分,其結(jié)構(gòu)與性質(zhì)會顯著影響蛋清蛋白在食品加工過程中的功能性質(zhì)。因此研究卵清蛋白在熱處理或美拉德反應(yīng)后的變化對了解蛋清蛋白在食品加工過程中的結(jié)構(gòu)與性質(zhì)的變化有重要意義。

 

卵清蛋白是一種優(yōu)質(zhì)蛋白質(zhì),占蛋清蛋白總量的54%-69%,卵清蛋白是典型的球蛋白,分子量為44.5kDa,屬含磷糖蛋白,含有四個自由巰基、385個氨基酸殘基[1]  。這些氨基酸殘基相互纏繞折疊形成具有高度二級結(jié)構(gòu)的球型結(jié)構(gòu),大部分為α-螺旋和β-折疊。卵清蛋白中心有1個二硫鍵和4個巰基,當(dāng)加熱時暴露出來,通過分子間相互作用使卵清蛋白膠體結(jié)構(gòu)變得更穩(wěn)定

【乳清蛋白lactalbumin

該納米載體取材綠色、設(shè)計巧妙,可在腫瘤部位依次針對腫瘤表面過表達(dá)酶、微酸和高谷胱甘肽(GSH)濃度等微環(huán)境實現(xiàn)三重響應(yīng)性釋藥(圖2)。作者首先通過定向酶解奶酪副產(chǎn)物-乳清蛋白獲得兩親性多肽,在水溶液中可自組裝形成30納米單分散膠束peptosome(PS),高效荷載疏水性抗癌藥物姜黃素Cur,解決了姜黃素水溶液難溶的問題,并降低其對正常組織的副作用。隨后他們對其表面修飾功能肽段PePm,其中Pe可靶向乳腺癌表面的ErB2受體,且兼具較好的抗癌轉(zhuǎn)移效果。此外,Pm片段可以根據(jù)腫瘤表面過表達(dá)金屬基質(zhì)蛋白酶MMP的特異性水解,通過交聯(lián)單個載體內(nèi)部自身巰基形成二硫鍵,使PS具有癌細(xì)胞高GSH濃度響應(yīng)釋藥的功能。他們將所構(gòu)建的載體PePm/PS/Cur經(jīng)尾靜脈注射到小鼠體內(nèi)后,經(jīng)EPR效應(yīng)和Pe的靶向效應(yīng)在腫瘤部位高度富集,載體表面Pe肽被MMP水解脫離,使Pe肽留在腫瘤細(xì)胞表面發(fā)揮抑瘤和抗轉(zhuǎn)移效果。PS/Cur繼續(xù)通過“質(zhì)子海綿”效應(yīng)從癌細(xì)胞溶酶體中逃逸,在細(xì)胞質(zhì)內(nèi)通過GSH響應(yīng)逐漸解體,釋放出PS/Cur內(nèi)荷載的姜黃素,發(fā)揮抑制腫瘤的作用。

【熒光標(biāo)記刀豆求蛋白A】:

刀豆球蛋白ACon A),又名刀豆凝集素、刀豆素,是一種植物血凝素,具有強力的促有絲分裂作用,有較好的促淋巴細(xì)胞轉(zhuǎn)化反應(yīng)的作用,其促淋巴細(xì)胞轉(zhuǎn)化最適濃度為40-100μg/ml,能沉淀肝糖原,凝集羊、馬、狗、兔、豬、大鼠、小鼠、豚鼠等動物及人紅細(xì)胞。還能選擇性激活抑制性T細(xì)胞(Ts)細(xì)胞,對調(diào)節(jié)機體免疫反應(yīng)具有重要作用。因此,通過使用Con A來活化病態(tài)(或老年)時的Ts細(xì)胞這一途徑,可望改觀一些自身免疫性疾病,甚或移植物排斥反應(yīng)或惡性腫瘤的防治前景。

 

刀豆蛋白(ConcanavalinACon A)是從刀豆(Jack bean)中提取出來的凝集素,能沉淀多種糖類,葡聚糖和果聚糖等很多其它多糖,以及免疫球蛋白和血型物質(zhì)等多種糖蛋白,還沉淀肺炎球菌多糖,并能凝集多種紅細(xì)胞。Con A 能和很多細(xì)菌和動物細(xì)胞反應(yīng),能區(qū)分某些正常和腫瘤細(xì)胞,能促進(jìn)細(xì)胞分裂(促有絲分裂作用)。Con A IV 用于淋巴下?lián)苻D(zhuǎn)化實驗,也被用來刺激T細(xì)胞產(chǎn)生IL-1 樣因子,還用于脾細(xì)胞增生,研究激活所必需的配體/受體作用。此外,尚區(qū)分正常分泌ACTH 的垂體細(xì)胞和分泌ACTH 的腺腫瘤細(xì)胞、用于研究糖基化在離子通道調(diào)節(jié)中的作用、用于糖蛋白的分離純化和多糖、糖蛋白糖鏈結(jié)構(gòu)的研究等。

Concanavalin A (con A) is a homotetrameric,26 kDa lectin derived from the jack bean, Canavalia ensiformis. Concanavalin Abinds to alpha-D glucose and alpha-D-mannose containing saccharides.Concanavalin A requires both Mn2+ and Ca2+ ions as a co-factor for binding.Since each con A has four binding sites, it can agglutinate cells such as redblood cells. Con A can also cause cell surface receptor aggregation which canbe mitogenic for some lymphocytes.

【熒光標(biāo)記親和素和鏈霉親和素】:

 

鏈霉親和素(streptavidin下稱SA)是與親和素(avidin下稱AV)有相似生物學(xué)特性的一種蛋白質(zhì),是streptomyces avidinii菌的分泌物,其分子量及結(jié)合生物素的能力與雞蛋清中的親和素相似,等電點6.0,非特異性結(jié)合遠(yuǎn)比親和素低。

 

鏈霉親和素是四聚體蛋白,大小為66KDa。一分子鏈霉親和素可以高度特異性地與四分子生物素結(jié)合,兩者之間的親和力極為強烈,鏈霉親和素-生物素復(fù)合物的解離常數(shù)處于10-14 mol/L 數(shù)量級,這一性質(zhì)常用于分子生物學(xué)用途。其中一條完整的SA肽鏈中有159個氨基酸殘基,分子量為16450

 

Avidin is a tetrameric protein commonlyisolated from egg whites. Avidin binds to biotin with strength approaching thatof a covalent bond. Disruption of the avidin-biotin bond occurs only underextraordinary conditions, such as pH 1.5 with 6 M guanidium hydrocholoride.Fluorescent avidin can bind to biotinylated antibodies used inimmunohistochemistry and immunocytochemistry. Avidin thiol can bind to goldsurfaces and act as a glue for biotinylated proteins. Avidin agarose is usefulfor efficient removal of biotinylated proteins from a liquid phase.

【熒光標(biāo)記和活化改性重組蛋白A】:

Fluorescein labeled Protein A is one of theRuixibio' premier fluorescent Protein A conjugates. Protein A is a 42 kDasurface protein originally found in the cell wall of Stapylococcus aureus.Protein A has strong binding affinity towards immunoglobulins and it has beenused for antibody identification, purification and many other biochemicalresearch uses. Fluorescein protein A is a green fluorescent protein A conjugatethat has excitation/emission wavelength at 495 nm/515 nm. All our protein Aconjugates were purified by size exclusion chromatography to removenon-conjugated molecules and ensure adequate applications both in-vitro andin-vivo.

產(chǎn)品介紹:

天然蛋白AProtein A)是一種發(fā)現(xiàn)于金黃色葡萄球菌的細(xì)胞壁表面蛋白,天然蛋白GProtein G)是一種分離自G型或C型鏈球菌屬的細(xì)胞表面蛋白,二者功能相似,主要通過與免疫球蛋白(Ig)的Fc區(qū)相互作用,可結(jié)合大多數(shù)哺乳動物的IgG。然而兩者結(jié)合特異性上有所不同(詳細(xì)見附錄1,蛋白A,G對不同物種Ig的結(jié)合能力總表),如蛋白G對人IgG3,大鼠IgG2a,綿羊IgG1具有很高的親和力,但蛋白A對這些Ig結(jié)合力很弱。蛋白A,蛋白G常共價偶聯(lián)在固體載體如瓊脂糖珠或丙烯酸樹脂小珠上,直接用以做免疫沉淀(IP)或者免疫共沉淀(Co-IP)來進(jìn)行蛋白功能相關(guān)研究,或者直接裝在層析柱上來純化單克隆或者多克隆抗體。

 

西安瑞禧生物科技有限公司使用的是基因改造后的蛋白A和蛋白G,不僅維持其本身的Ig親和特性,同時也去除了天然蛋白本身的非主要結(jié)合域以降低非特異性結(jié)合。本品是同時共價偶聯(lián)了蛋白A和蛋白G的瓊脂糖珠,比單獨的蛋白A或者蛋白G都有更廣的結(jié)合范圍,適合于所有蛋白A瓊脂糖珠和蛋白G瓊脂糖珠單獨可以結(jié)合的Ig,實用性更高。本品為50%的懸液,1 ml Protein A/G 瓊脂糖珠可結(jié)合約25 mgIgG

Protein G is an immunoglobulin bindingprotein in Streptococci. Protein G binds to the Fc region of antibodies andthereby leaves the Fab region available to still bind antigen. Our recombinantversion has been engineered to lack the albumin binding domains and has a 6xhistidine tag at its N-terminus.

Protein AProtein G產(chǎn)品應(yīng)用:

重組蛋白A的C末端含有半胱氨酸,可定向偶聯(lián)到固體分離介質(zhì)上,用于單克隆抗體或多克隆抗體的純化??山Y(jié)合大多數(shù)人類和小鼠免疫球蛋白子類(如。,人類IgG1 IgG2IgG4;鼠標(biāo)IgG2 IgG2a,IgG2b IgG3),還結(jié)合牛,幾內(nèi)亞豬,倉鼠,房子,豬,兔子總免疫球蛋白的形式。重組蛋白A可與多種報告分子(熒光染料、酶標(biāo)記、生物素、膠體金、放射性標(biāo)記等)偶聯(lián)并不影響其與抗體結(jié)合的活性。這些偶聯(lián)后的蛋白A衍生物可用于Western-blot、ELISA、免疫組化等的抗體檢測。

 

 

Greenfluorescent protein (GFP)綠色熒光蛋白】:

Green fluorescent protein (GFP) is a 27 kDaprotein derived from Aequorea victoria. It absorbs light in the ultraviolet toblue range and emits green (peak emission 510 nm). GFP is a common reportermolecule for protein expression and visualization of intracellular processes.We produce this protein in E.coli.
